چکیده

Introduction Neutrophils are the primary defense of the host against pathogens. They remain in resting state in the circulation of the healthy individuals. When encountering cytokine or chemokine signals caused by pathogen products, neutrophils are activated and mobilized to the infected sites. The life span of neutrophils which have an essential role in terms of pathogenesis of autoinflammatory diseases are very short. In consideration of this issue, it is restricted to study neutrophil migration in vitro. Hence, we developed a new approach on cell migration assays and neutrophil priming for long lasting research.

Investigation of the inflammatory cell migration process in familial Mediterranean fever

Introduction Familial Mediterranean fever (FMF) is one of the most common autoinflammatory disorders and is characterized by episodic attacks of fever, along with inflammation. FMF pathogenesis is associated with various mutations in the MEFV gene, which encodes pyrin. Pyrin is expressed predominantly in neutrophils that have an important role in the innate immune response. Several proteins rel...

Expression of pro-inflammatory genes in lesions, spleens and blood neutrophils after burn injuries in mice treated with silver sulfodiazine

Objective(s): It is now supposed that cytokines released during the burn injuries have a great impact on the immunological and pathological responses after the burn. The main objective of this study was to investigate the expression of some pro-inflammatory genes in the wound, spleen and blood neutrophils during the healing process of burn wounds in a murine model. Materials and Methods: The ex...

Drift-Diffusion Analysis of Neutrophil Migration during Inflammation Resolution in a Zebrafish Model

Neutrophils must be removed from inflammatory sites for inflammation to resolve. Recent work in zebrafish has shown neutrophils can migrate away from inflammatory sites, as well as die in situ. The signals regulating the process of reverse migration are of considerable interest, but remain unknown. We wished to study the behaviour of neutrophils during reverse migration, to see whether they mov...

Impaired immune tolerance to Porphyromonas gingivalis lipopolysaccharide promotes neutrophil migration and decreased apoptosis.

Periodontitis, a chronic inflammatory disease of the tissues supporting the teeth, is characterized by an exaggerated host immune and inflammatory response to periopathogenic bacteria. Toll-like receptor activation, cytokine network induction, and accumulation of neutrophils at the site of inflammation are important in the host defense against infection. At the same time, induction of immune to...

Inhibitors of neutrophil recruitment identified using transgenic zebrafish to screen a natural product library

Cell migration is fundamental to the inflammatory response, but uncontrolled cell migration and excess recruitment of neutrophils and other leukocytes can cause damage to the tissue. Here we describe the use of an in vivo model - the Tg(mpx:GFP)(i114) zebrafish line, in which neutrophils are labelled by green fluorescent protein (GFP) - to screen a natural product library for compounds that can...
